How It Works: SureSilencing™ siRNA Arrays

The SureSilencing siRNA Array is the latest technological innovation for conducting functional studies on your pathway of interest using RNA interference (RNAi). The SureSilencing siRNA Arrays may be used for accurate pathway analysis, the functional analysis of novel genes, and drug target, enhancer, or inhibitor validation in stable transformed human cell lines. The siRNA Array is a set of siRNA pairs validated to knock down by RNA interference 42 pathway or disease focused genes. The siRNA is arrayed in duplicates, along with appropriate RNAi and assay controls, in a 96-well cell culture plate compatible with any cell-based chemiluminescent, colorimetric, or fluorescent assay. Simply add your medium and transfection reagent to resuspend the siRNA, and add your cells for reverse transfection. Then, perform your cell-based phenotype assay directly in the array.   1. Add medium and transfection agent to wells pre-coated with siRNA.
  2. Plate trypsinized cells in suspension into the wells.
  3. Analyze phenotype using cell-based assays.

The reverse transfection method produces equivalent or improved transfection efficiency over standard pre-plated methods* and saves an entire day in the process. The simplicity and reliability of the siRNA Array allows you to quickly analyze gene function and mechanism of action in your pathways of interest.

Layout and Controls: The duplicate siRNA pairs are arrayed in adjacent wells for easy readout of the phenotype assay results. The control wells include negative control siRNA, mock transfection controls, fluorescently labeled siRNA for monitoring transfection efficiency, and phenotype assay background controls.
