| Description |
| RNASafer is an aqueous, non-toxic tissue
or cell storage reagent that can rapidly stabilize and protect cellular
RNA in situ in unfrozen or frozen samples. RNA extractions can be made
from cell or tissue suspensions stored in RNASafer at -20 °C for more
than one year. We recommend storing only fresh, previously unfrozen tissue
or cell samples in RNASafer. Sufficient volumes of RNASafer are necessary
to effectively preserve RNA in tissues. You can use any RNA isolation kit
to extract RNA from samples stored in RNASafer.

Kit Contents: One 100ml bottle containing RNASafer™ RNA
Stabilization Reagent.
Storage Conditions:
The product is shipped at ambient temperature and may be stored at room
temperature. The product is stable for at least 6 months from the date
received. If precipitation occurs, heat the solution to 37 °C to
re-dissolve.

| Brief Protocol |
All steps during sample preparation and RNA isolation should be carried
out as quickly as possible. Crystals found in the RNASafer suspension
after storage will not affect RNA isolation.
- Tissue Samples:
- Excise tissue samples from the animal quickly (within 2 minutes),
cutting it into slices less than 0.4 cm thick.
- Immediately, add a sufficient volume of RNASafer to completely
submerge the slices.
Note: Typically ~12 µl of RNASafer per mg of tissue is sufficient to
preserve the sample. The amount varies according to tissue type and
container volume. It is essential that all tissue be covered in
RNASafer to effectively preserve RNA. An insufficient amount will
allow RNA degradation.
- Store tissue samples in RNASafer at -20 °C.
- Cell Samples:
- Pellet up to one million cells at 5000 x g. Discard culture broth.
- Gently suspend cells in 50 µl of PBS.
- Add 500 µl of RNASafer. Store at -20 °C.
- Homogenization of samples stored in RNASafer:
- Tissue Samples:
- Remove tissue from RNASafer using forceps or other means
- Homogenize tissue in lysis buffer or Trizol and then isolate RNA.
- Cell Samples
- Dilute RNASafer suspension two-fold with PBS.
- Immediately, pellet cells at 5000 x g. Remove supernatant.
- Add cell lysis buffer and then isolate RNA.
- Cell or tissue sample can be stored in RNASafer for up to:
1 day at 37 °C
9 days at 25 °C;
1 month or more at 4 °C
1 year or more at -20 °C or -80 °C
