| Description |
| The XpressRef™ Universal Total RNA samples contain the broadest possible representation of genes from a specific organism. These RNA samples can serve as an internal normalization control for two-color microarray systems and serve as a troubleshooting tool for single color microarray systems, in particular, the Oligo GEArray®. These RNA samples are also useful to troubleshoot real-time or semi-quantitative RT-PCR gene expression analyses when the gene of interest is difficult to detect or to optimize real-time RT-PCR gene expression assays by allowing the determination of amplification efficiencies when experimental RNA samples are precious. Two aliquots of 100 µg RNA each at 1.0 mg/ml are provided in this kit, enough for many array or RT-PCR experiments.

| Brief Protocol: |
A. For use with SABiosciences Oligo GEArray® System:
- Follow the instructions provided in the Oligo GEArray® System and the TrueLabeling-AMP™ 2.0 kit User Manuals for all steps of this protocol.
- Generate labeled target from 3 µl (3 µg) of the XpressRef™ Universal Total RNA using the TrueLabeling-AMP™ 2.0 Kit (GA-030) at the same time as your experimental RNA samples.
- Hybridize the labeled targets to separate Oligo GEArrays® with the same catalog number.
- Detect the labeled target hybridized to the array.
- A number of spots should appear on the array image if the probe synthesis reaction, hybridization, washing and detection steps were all successfully performed.
B. For use with other microarray systems:
Follow the recommendations provided by the microarray
manufacturer for using universal RNA.
C. For use with the SABiosciences real-time RT²Profiler™ PCR Array System or RT² Real-Time™ PCR Primer Sets and RT² Real-Time™ master mixes:
- Follow the instructions provided in the RT²Profiler™ PCR Array System and the RT² Gene Expression Assays User Manuals for all steps of this protocol.
- Use 3 µl (3 µg) of the XpressRef™ Universal Total RNA with the RT² PCR Array First Strand Kit (C-02) protocol.
- For the primer sets, prepare ten-fold serial dilutions of the cDNA template for separate real-time reactions to generate your calibration curve.
- For the RT²Profiler™ PCR Array System, a number of wells should provide threshold cycle (Ct) values < 35 if the cDNA synthesis and real-time PCR cycling conditions were all successfully performed.
