RT² Profiler PCR Arrays and RT² qPCR Primer Assays:
Verify that nuclear factor and/or RNA Polymerase II enrichment at your promoter of interest alters transcript levels of the relevant gene and/or other genes in the same biological pathway using real-time RT-PCR. The RT² qPCR Primer Assays are designed to analyze the transcript levels of individual genes, while the RT² Profiler™ PCR Arrays profile a panel of pathway-focused or disease-specific mRNAs. Both methods are optimized for high performance SYBR Green based qRT-PCR detection.

SureSilencing shRNA Plasmids:
Test whether other related nuclear or transcription factors are in the same complex as your ChIP target factor by knocking down the expression of another factor of interest and repeating your ChIP-qPCR experiment to test whether the target factor is still enriched at the promoter region of interest. Confirm the activation of the signal transduction pathway upstream of your nuclear or transcription factor of interest by monitoring the phosphorylation of the pathway's regulators using a cell-based in vitro assay. Or verify the induction of cytokines at the protein level caused by the observed enrichment of nuclear factors at their promoter using standard sandwich ELISA kits.
