What Is the Oligo GEArray?

The Oligo GEArray is our latest generation of pathway-focused DNA microarray. The nylon membrane array matrix is a permeable support with a high DNA binding capacity. The production process employs a reproducible non-contact printing technology to prevent deformations in the membrane. The carefully designed 60-mer oligonucleotide probes printed on each microarray minimizes any cross-hybridization between spots on the same array despite the representation of closely related members of the same gene families. An advanced target synthesis and labeling protocol provides un-biased linear RNA amplification in a simple one-tube protocol. The biotinylated cRNA target and the carefully designed oligonucleotides on the array correspond to the same 3'-biased gene-specific sequences permitting efficient and specific hybridization. The Oligo GEArray is also optimized for the popular chemiluminescent detection method, which allows the use of either X-ray film or a CCD-camera imaging system for image acquisition.

Two different formats and three different protocols available:
We offer two different formats for processing the Oligo GEArrays, the HybPlate and HybTube formats. In the HybTube format, the Oligo GEArray membranes are shipped, stored, and used in their own disposable plastic tubes for adaptability to most standard rotisserie-type or roller bottle hybridization ovens. In the innovative HybPlate format, the Oligo GEArray membranes are shipped and stored in an ExpressPak box and are conveniently used in 8-well Multi-chamber HybPlates for easier handling. The microarrays and plates can be used in any standard 60 ºC incubator with a static two-day protocol or in a specialized but inexpensive Thermoshaker (for only ~$2000) for a one-day agitation protocol for even faster and more high-throughput analyses. The overall methodology and performance of all three protocols and two formats are comparable. Simply choose a protocol based on the ease-of-use that you require and the equipment available in your laboratory.

How It Works:

Isolate total RNA from your experimental biological source material using an appropriate kit and method. Enzymatically convert the RNA into a biotinylated cRNA target with the TrueLabeling-AMP™ 2.0 Kit in a standard thermal cycler. Hybridize the labeled target to the GEArray microarrays in a HybPlate and either an incubator or the Thermoshaker or hybridize in a HybTube in a standard hybridization oven. Detect the microarray with the Chemiluminescent Detection Kit reagents and either a Chemi-Doc imaging system (CCD camera) or X-ray film. Acquire and analyze the data with our web-based software package.
